Product Description

 

This product adopts a simple and easy way. The formula is optimized to ensure that it does not destroy white blood cells but can fully lyse and remove red blood cells. It is suitable for fresh blood of humans, mice and other mammals. Under normal circumstances, more than 80% of all white blood cells in blood samples can be obtained and the biological activity of white blood cells can be fully guaranteed.

The main component of this product is ammonium chloride. It is a ready-to-use solution that can quickly and effectively lyse red blood cells and enrich and separate white blood cells. The obtained white blood cells have good biological activity and can be directly used in downstream experiments such as flow cytometry, nucleic acid or protein extraction, and PCR.


Transportation and storage methods

 

Transport with wet ice. Store at 4℃, valid for one year. Store at room temperature, valid for three months.

 

Precautions

 

1) This reagent is suitable for separating leukocytes from 1μL to 10mL of blood samples. The obtained leukocytes can be used for downstream experiments.

2) To extract the best nucleic acid or protein, please use fresh blood or blood that has been stored for less than 7 days. If blood clots are formed, remove them. Try to use anticoagulant tubes to collect samples.

3) Normally, blood samples and red blood cell lysis buffer are lysed in a ratio of 1:3. For some special diseases such as leukemia, polycythemia, etc., it is recommended to use a ratio of 1:4 or higher to ensure the results.

4) The main ingredient of this product is ammonium chloride, which cannot guarantee the effective lysis of nucleated red blood cells, such as those in the blood of birds, reptiles, fish, etc.

5) This product can also be used for the lysis of red blood cells in mammalian tissues.

Procedure

 

Before use, equilibrate the cryopreserved red blood cell lysis buffer to room temperature. Then add 3 times the volume of red blood cell lysis buffer to the blood sample (for example, add 900 μL of red blood cell lysis buffer to 300 μL of blood), invert and mix, leave at room temperature for 5 minutes, invert and mix several times, centrifuge at 10000rpm for 1 minute (if the maximum speed of the centrifuge does not allow, centrifuge at 3000rpm for 5 minutes), and remove the supernatant. Wash once with an appropriate buffer (such as PBS, pH 7.2~7.4). After centrifugation, leave the white blood cell pellet for subsequent operations.
