【Detailed description of yeast culture medium】

 

      The first generation medium for yeast genetics research is a formula that was widely used in early laboratories for yeast culture. It requires mixing an amino acid mixture/selection of nutritional deficiency with a yeast nitrogen source (Yeast Nitrogen Base, YNB), which is divided into ammonium sulfate-containing and ammonium sulfate-free. This often causes some novices to make mistakes when preparing it. In fact, in addition to being more troublesome to prepare, the more critical point is that the amino acid components of its traditional formula are not suitable for some special prototrophic yeasts or genetically defective yeasts. This is also the reason why some laboratories often encounter problems when using the first generation medium without knowing why. Therefore, most laboratories doing yeast genetics research are now using the second generation medium improved according to the principle of defect pairing complementation enhancement (Complementation test), which can be directly prepared and sterilized by adding water.

     First-generation products and second-generation products: Amino acid mixture is a first-generation product. It is generally not recommended for customers to continue using first-generation products. Currently, most universities and the Chinese Academy of Sciences system are using our second-generation products.

There are two types of yeast selective nutrient deficiency media:

 

      (1) A type of culture medium, also known as the first generation culture medium, is actually an amino acid mixture (such as the 10 g Do supplement produced by Cl***tech). In fact, it cannot be called a culture medium because it requires the addition of yeast nitrogen source and amino acid mixture before it can be used. On the one hand, it is more troublesome to prepare, and on the other hand, it is easy to make mistakes.

      (2) Type II culture medium is the second generation culture medium. One of its main features is that it is very convenient to prepare and use. No need to add Minimal SD Base

      Another notable feature of our second-generation culture medium is that it optimizes the nutritional elements and enhances the combination of nutrients. It is not only stable and has a long shelf life, but also has a wider range of applications. It is suitable for all yeasts used in genetic research, including those with special mutant strains. Although Cl***tech currently has a second-generation culture medium, it is not universal for yeast. Our second-generation culture medium is not only easy to use, but more importantly, it has been enhanced and improved in nutritional components, which is particularly suitable for yeast with certain genetic defects. This is our original creation.

      In fact, the second-generation culture medium was first developed by our company. Six years after our second-generation culture medium was launched, Cl***tech (now acquired by Ta**ra) began to produce this second-generation culture medium. (Cl***tech only had the first-generation product before that, which was a 10-gram bottle of Do supplement, also known as Dropout, which required the addition of Minimal SD base at the same time).

 

      Relevant laboratories of several institutes of the Chinese Academy of Sciences and several laboratories of Peking University used Cl***tech's second-generation products and similar cheap products from other domestic companies, and found that yeast growth was delayed and discolored. Later, they switched to our products to solve the problem. Objectively speaking, Cl***tech's second-generation products are no problem for many types of yeast, but may not work for some special genotypes of yeast. Our second-generation culture medium belongs to the [universal enhanced type], which is not only applicable to a wide range of applications, but also has unique effects.
